utility of myelin basic protein as an early prognostic biomarker in multiple sclerosis

conclusions molecular assays in the early stages of the disease could help in elucidating the effectiveness of the mbp as a prognostic factor in ms. methods this study is convergence of three arms including in silico and in vitro (bioinformatics) with the in vivo (experimental). a novel methodology combining molecular techniques was used to confirm the antigenic properties of mbp and study its efficiency in increasing the susceptibility to ms. one hundred eighty ms patients and healthy subjects were recruited for the study from jan 2013 to feb 2016 in iran. age and sex-matched healthy volunteers and patients were analyzed using various quantitative and qualitative molecular laboratory techniques. peripheral blood mononuclear cells (pbmcs) and plasma was used for the retrieval of mbp and igg assay, respectively. background myelin basic protein (mbp), a crucial neuro-autoantigen involved in the maintenance of the myelin sheath, is one of the biomarkers of therapeutic response in multiple sclerosis (ms). results the optimum concentration of the mbp epitope for the immune system to react and facilitate prognostication was found to be 50 and 150 µg/ml in ms patients and healthy individuals, respectively (p < 0.0001****). combined results from elisa and real-time pcr showed that the total igg and the ratio of gene expression for candidate human mbp epitope was higher in ms patients in all the three groups compared to that in healthy controls p < 0.0001****). objectives the study examines prognostic biomarker and molecular mimicry hypothesis ms etiology by mbp.

Serum and Saliva Myelin Basic Protein as Multiple Sclerosis Biomarker
Objective: Multiple sclerosis (MS) is presented with motor and sensory function loss. It is caused by demyelination and following axonal lesion. As myelin basic protein (MBP) is one of the key elements of the myelin cover, we examined the level of MBP in serum, stimulated, and unstimulated saliva as a suitable biomarker for detecting MS. Methods: A case-control study was performed in 29 health...

Combination of Myelin Basic Protein Gene Polymorphisms with HLA-DRB1*1501 in Iranian Patients with Multiple Sclerosis
Background: Multiple sclerosis (MS), as a multifactorial autoimmune disease with complex genetic basis, causes demyelination in the central nervous system via cytokine responses to myelin antigens. Myelin basic protein (MBP) is the main protein component of the myelin sheath. HLA-DRB (human leukocyte antigen-DR beta) alleles, particularly HLA-DRB1*1501, may be of significance in the pathogenesi...
full textCerebrospinal fluid myelin basic protein as a prognostic biomarker in dogs with thoracolumbar intervertebral disk herniation.
BACKGROUND Release of myelin basic protein (MBP) into the cerebrospinal fluid (CSF) is associated with active demyelination and correlates with outcome in various neurological diseases. HYPOTHESIS/OBJECTIVES To describe associations among CSF MBP concentration, initial neurological dysfunction, and long-term ambulatory outcome in dogs with acute thoracolumbar intervertebral disk herniation (I...
full textEffect of methylprednisolone on CSF IgG parameters, myelin basic protein and anti-myelin basic protein in multiple sclerosis exacerbations.
Clinical exacerbations of multiple sclerosis (MS) are characterized by elevated levels of cerebrospinal fluid (CSF) myelin basic protein (MBP). The purposes of this study were to determine whether anti-MBP antibodies are present in increased titer in CSF of MS patients with exacerbations, and whether they can be suppressed by the administration of immunosuppressive dosages of methylprednisolone...
full textMyelin basic protein, oligoclonal bands, and IgG in cerebrospinal fluid as indicators of multiple sclerosis.
There currently are three clinical laboratory procedures for use with cerebrospinal fluid that assist in the diagnosis of multiple sclerosis: measurement of myelin basic protein and IgG, and demonstration of an oligoclonal band. We compared characteristics of these procedures, using CSF samples from 166 patients identified as having (54 patients) or not having (112 patients) multiple sclerosis....
